Will the rack take 27.5+ size tires?

yes, It'll fit a fat tire 27.5 along with a 29er. I'm very happy with this rack.

I own this product and use it for my Chevy S-10 small pickup. It works well, is sturdy and easy to use, and I think well designed. I don’t know how it might work in a trailer but I think it would be adaptable.

Will it fit a 27.5 X 3.0 and a 26X4.9 wheel?

it might. I put a roll cover over the back of my f150 and could not use the bike rack along with the cover so I sold the bike rack. It's not practical if you want a bed cover for the back of your truck.

how does this rack lock to the truck bed?

The rack secures to the bed with a bumper that is secured with large wing nuts. The rack also comes with a small cable lock.
