Q: Will this bulb work pull string switch

Mine work off a conventional switch outside the bathroom so as long as a pull switch is properly wired in yes it should work as the string operates the switch not the lamp. If the switch is working the lamp should come on.

Q: My original bulb is a 16W. Can i replace it with a 28W for increased brightness?

Yes I would think that would be okay as long as it is a Square 4 Pin Bulb
